Overview

This is a multi-agent combination regimen consisting of six drugs—cyclophosphamide, doxorubicin, fluorouracil, methotrexate, conjugated estrogens, and tamoxifen—used in the treatment of advanced or metastatic breast cancer. Each component has a distinct mechanism of action: - Cyclophosphamide is an alkylating agent that crosslinks DNA strands to inhibit cell division. - Doxorubicin is an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II. - Fluorouracil (5-FU) is a pyrimidine analog that inhibits thymidylate synthase and disrupts DNA synthesis. - Methotrexate is an antimetabolite that inhibits dihydrofolate reductase, blocking folic acid metabolism required for nucleotide synthesis. - Conjugated estrogens are mixtures of estrogen hormones used for hormone modulation. - Tamoxifen is a selective estrogen receptor modulator (SERM) acting as an antagonist on breast tissue. This combination represents both cytotoxic chemotherapy and hormonal therapy approaches. It has been studied primarily in the context of advanced or metastatic breast cancer to maximize antitumor efficacy by targeting multiple pathways involved in tumor growth and proliferation[1][3].
