Overview

This is a combination regimen consisting of three drugs: - Cyclophosphamide, an alkylating agent that cross-links DNA strands and inhibits DNA synthesis, leading to cell death. - Doxorubicin, an anthracycline antibiotic that intercalates into DNA and inhibits topoisomerase II, causing double-strand breaks and preventing replication. - Pembrolizumab, a monoclonal antibody immune checkpoint inhibitor targeting the programmed cell death protein 1 (PD-1) receptor on T cells. By blocking PD-1 interaction with its ligands (PD-L1/PD-L2), pembrolizumab enhances T-cell-mediated anti-tumor immune responses. Cyclophosphamide plus doxorubicin is a well-established chemotherapy backbone for breast cancer and other malignancies[2][3][4]. Pembrolizumab is approved for multiple cancers as monotherapy or in combination with chemotherapy[6][8]. The triple combination is under investigation in clinical trials for various solid tumors to leverage both cytotoxic effects of chemotherapy and immunomodulatory effects of checkpoint inhibition.
