This is a combination chemotherapy regimen consisting of four agents: - **Cyclophosphamide** (an alkylating agent), - **Doxorubicin** (an anthracycline antibiotic), - **Uracil** (a nucleobase and dihydropyrimidine dehydrogenase inhibitor), and - **Tegafur** (a prodrug of 5-fluorouracil). The combination is sometimes referred to as "CAUT" in the literature. It has been studied primarily for the treatment of advanced or recurrent breast cancer. The rationale for this regimen is to combine multiple mechanisms of cytotoxicity—DNA crosslinking by cyclophosphamide, DNA intercalation and topoisomerase II inhibition by doxorubicin, and antimetabolite activity via 5-FU generated from tegafur with uracil enhancing its bioavailability—while aiming to reduce toxicity compared to traditional regimens using intravenous 5-FU. Clinical studies have shown that this combination can achieve high response rates with relatively mild adverse reactions in advanced breast cancer patients[5][6].
