Overview

This is a combination therapy consisting of cyclophosphamide (an alkylating agent), doxorubicin (an anthracycline topoisomerase inhibitor), and veliparib (an oral poly(ADP-ribose) polymerase (PARP) inhibitor). Veliparib potentiates the effects of alkylating agents and topoisomerase inhibitors in preclinical tumor models. The combination has been studied in phase I clinical trials for advanced malignancies, particularly in metastatic breast cancer. The mechanism involves veliparib delaying repair of DNA damage induced by the chemotherapeutic agents, with increased DNA damage observed through markers like γH2AX in circulating tumor cells.
