Overview

Cyclophosphamide + exemestane is a combination therapy used primarily in the treatment of hormone receptor–positive breast cancer, particularly in postmenopausal women. Cyclophosphamide is an alkylating agent that damages DNA and inhibits cancer cell replication, while exemestane is an aromatase inhibitor that blocks estrogen synthesis, reducing hormone-driven tumor growth. This combination has been studied as a neoadjuvant (pre-surgical) or metastatic therapy for patients who do not respond adequately to endocrine monotherapy with exemestane alone. The regimen typically involves daily oral administration of both drugs (exemestane 25 mg and cyclophosphamide 50 mg). Clinical studies have shown this combination can be beneficial for patients with limited response to initial endocrine therapy, offering improved clinical response rates and manageable safety profiles[1][2][3][4].
