Overview

This is a combination chemotherapy regimen consisting of three drugs: - **Cyclophosphamide** is an alkylating agent that interferes with DNA replication and RNA transcription by cross-linking DNA strands, leading to cell death. - **Fluorouracil (5-FU)** is an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis and function. - **Leucovorin (folinic acid)** enhances the cytotoxic effects of 5-fluorouracil by stabilizing the binding of 5-FU's active metabolite to thymidylate synthase. This combination has been studied in various cancers including advanced pancreatic cancer and metastatic breast cancer. Leucovorin acts as a biochemical modulator to increase the efficacy of 5-fluorouracil. Cyclophosphamide may further enhance this effect[1][2]. The regimen aims to maximize antitumor activity while managing toxicity.
