This is a combination chemotherapy regimen consisting of four agents: - Cyclophosphamide, an alkylating agent that interferes with DNA replication and cell division. - Fluorouracil (5-fluorouracil or 5-FU), an antimetabolite that inhibits thymidylate synthase, disrupting DNA synthesis. - Medroxyprogesterone acetate, a synthetic progestin hormone with anti-estrogenic effects used in some hormone-sensitive cancers. - Methotrexate, an antimetabolite and antifolate agent that inhibits dihydrofolate reductase, blocking DNA synthesis and cell proliferation. This combination has been studied for the treatment of recurrent or metastatic endometrial carcinoma. The regimen aims to combine cytotoxic effects on rapidly dividing tumor cells (via cyclophosphamide, fluorouracil, methotrexate) with hormonal modulation (via medroxyprogesterone acetate). Clinical studies have shown this combination can achieve objective responses in certain advanced cancers such as endometrial carcinoma[4].
