This combination regimen consists of five drugs—cyclophosphamide, fluorouracil (5-FU), methotrexate, prednisone, and tamoxifen—used together as adjuvant chemohormonal therapy primarily for breast cancer. Cyclophosphamide is an alkylating agent that crosslinks DNA; fluorouracil is a pyrimidine analog that inhibits thymidylate synthase; methotrexate is an antimetabolite inhibiting dihydrofolate reductase; prednisone is a synthetic glucocorticoid with anti-inflammatory and immunosuppressive effects; and tamoxifen is a selective estrogen receptor modulator (SERM) that antagonizes estrogen receptors in breast tissue. The combination aims to target cancer cells through multiple mechanisms including cytotoxicity and hormonal modulation. Clinical studies have evaluated this regimen in node-positive or high-risk early-stage breast cancer patients to assess whether the addition of prednisone and/or tamoxifen enhances outcomes compared to standard regimens[1][5]. Toxicity increases with the addition of prednisone and tamoxifen[1].
