Cyclophosphamide + glucocorticoids is a combination therapy consisting of the alkylating agent cyclophosphamide and one or more drugs from the glucocorticoid class (such as prednisone, methylprednisolone, or dexamethasone). This regimen is used primarily for its potent immunosuppressive and anti-inflammatory effects in severe autoimmune diseases (including membranous nephropathy, IgA nephropathy, Graves’ ophthalmopathy) and certain cancers. Cyclophosphamide acts by cross-linking DNA strands via its active metabolite phosphoramide mustard, leading to cell death—especially in rapidly dividing cells such as lymphocytes. Glucocorticoids suppress immune responses by inhibiting cytokine production and lymphocyte proliferation. The combination enhances efficacy in controlling disease activity but increases the risk of adverse effects such as infections and metabolic complications[3][5][8].
