Overview

This is a combination therapy consisting of three drugs—cyclophosphamide (an alkylating agent), letrozole (an aromatase inhibitor), and sorafenib (a multi-kinase inhibitor)—used primarily in the treatment of primary breast cancer. The regimen is designed to combine endocrine therapy with metronomic chemotherapy and targeted antiangiogenic therapy. Cyclophosphamide is administered at low, continuous doses ("metronomic" scheduling) to target tumor vasculature and reduce toxicity. Letrozole blocks estrogen synthesis by inhibiting aromatase, which is effective in postmenopausal women with hormone receptor-positive breast cancer. Sorafenib inhibits multiple kinases involved in tumor growth and angiogenesis[1][2][6]. The combination has shown clinical activity and tolerability in early-phase trials for primary breast cancer.
