Overview

CDD + bortezomib is an investigational combination chemotherapy regimen consisting of cyclophosphamide, liposomal doxorubicin, dexamethasone (collectively known as CDD), and the proteasome inhibitor bortezomib. This regimen is primarily being evaluated in Phase 3 clinical trials sponsored by Beijing Chao Yang Hospital for the treatment of patients with relapsed or refractory multiple myeloma who have developed extramedullary plasmacytoma (EMP). The combination utilizes a multi-targeted approach: bortezomib disrupts protein homeostasis by inhibiting the 26S proteasome; cyclophosphamide acts as a DNA alkylating agent; liposomal doxorubicin inhibits DNA topoisomerase II and intercalates DNA; and dexamethasone acts as a glucocorticoid receptor agonist to induce apoptosis in malignant plasma cells. This synergistic combination is designed to overcome treatment resistance and target malignant cells both within the bone marrow and in extramedullary sites.
