cyclophosphamide + megestrol acetate is a combination therapy used in oncology, particularly in the treatment of metastatic breast cancer and other advanced malignancies. Cyclophosphamide is an alkylating agent that works by interfering with DNA replication and cell division, leading to cytotoxic effects on rapidly dividing cells. Megestrol acetate is a synthetic progestin with hormonal activity; its antitumor mechanism may involve suppression of luteinizing hormone via inhibition of pituitary function and possible modulation of inflammatory cytokines. The combination has been studied in phase II trials as a metronomic regimen—low-dose, frequent administration—to reduce toxicity while maintaining efficacy. This approach aims to target both tumor cells directly (via cyclophosphamide) and hormone-sensitive pathways or supportive care needs (via megestrol acetate). The regimen has shown activity in pretreated patients with metastatic breast cancer, including those with hormone receptor-positive disease.[1][2][3]
