cyclophosphamide + mitoxantrone

Cyclophosphamide + mitoxantrone is a combination chemotherapy regimen consisting of two small molecule antineoplastic agents. Cyclophosphamide is an alkylating agent that interferes with DNA replication and RNA transcription, leading to cell death. Mitoxantrone is an anthracenedione derivative that intercalates into DNA, causing crosslinks and strand breaks, and inhibits topoisomerase II, resulting in cytotoxicity. This combination has been studied primarily for the treatment of advanced breast cancer and various lymphomas, as well as in salvage regimens for acute myeloid leukemia (AML). The regimen has demonstrated efficacy with manageable toxicity profiles; common adverse effects include granulocytopenia, alopecia, nausea, vomiting (mainly from cyclophosphamide), while mitoxantrone appears to have less cardiac toxicity compared to other anthracyclines[1][7][8].
