Overview

This refers to a dose-dense adjuvant chemotherapy regimen developed and evaluated by the University of Nebraska Medical Center for the treatment of Stage I-II breast cancer. The regimen consists of cyclophosphamide, an alkylating agent that cross-links DNA to prevent replication, and paclitaxel, a taxane that stabilizes microtubules to inhibit cell division. For patients with HER2-positive disease, the regimen includes the monoclonal antibody trastuzumab, which targets the human epidermal growth factor receptor 2 (HER2). The treatment is typically administered every two weeks for six cycles (dose-dense schedule), followed by trastuzumab maintenance for HER2-positive patients. This non-anthracycline approach was designed to provide an effective adjuvant option while minimizing the long-term cardiac and hematologic toxicities often associated with anthracycline-based regimens.
