Overview

Cyclophosphamide + sirolimus is a combination therapy consisting of two small molecule drugs. Cyclophosphamide is an alkylating agent that crosslinks DNA, leading to cell death, and is widely used as a chemotherapeutic and immunosuppressive agent. Sirolimus (also known as rapamycin) is an inhibitor of the mammalian target of rapamycin (mTOR), blocking cell proliferation and angiogenesis by inhibiting mTOR signaling pathways. This combination has been investigated for synergistic anti-tumor activity in sarcoma, where activation of the mTOR pathway contributes to tumor growth[1]. It has also been studied in post-transplant settings to reduce graft-versus-host disease by modulating immune responses[3][5]. The regimen aims to leverage both cytotoxic and cytostatic effects for improved disease control.
