Overview

This is a combination therapy consisting of cyclosporin A (CsA), an immunosuppressant, and recombinant human thrombopoietin (rhTPO), a biologic agent that stimulates platelet production. Cyclosporin A acts primarily by inhibiting calcineurin, thereby suppressing T-cell activation and immune response. Recombinant human thrombopoietin mimics endogenous thrombopoietin to stimulate the proliferation and differentiation of megakaryocytes from bone marrow progenitors, leading to increased platelet production. This combination has been investigated as a second-line treatment for corticosteroid-resistant primary immune thrombocytopenia (ITP) and non-severe aplastic anemia (NSAA). Clinical studies indicate that the combination can accelerate platelet recovery, reduce relapse rates compared to monotherapy with rhTPO or CsA alone, and is generally well tolerated with mild adverse events[1][2][5][6].
