Cyclosporine is an immunosuppressive agent used to prevent organ rejection in transplant patients and to treat severe active rheumatoid arthritis and plaque psoriasis. It works by inhibiting calcineurin, thereby suppressing T-cell activation and the immune response[2]. Berotralstat (BCX7353) is a synthetic, oral, once-daily small molecule that selectively inhibits plasma kallikrein. It is developed for the prevention and treatment of hereditary angioedema (HAE) attacks by blocking the kallikrein–kinin system pathway involved in bradykinin production[5][6]. The combination "cyclosporine + BCX7353" refers to co-administration studied primarily for drug-drug interaction purposes rather than as a therapeutic fixed-dose combination; specifically, cyclosporine has been evaluated for its effect on the pharmacokinetics of berotralstat[7].
