Cyclosporine + daclizumab is a combination of two immunosuppressive agents used primarily to prevent acute rejection in organ transplantation, particularly kidney transplants. Cyclosporine is a calcineurin inhibitor that suppresses T cell activation by inhibiting the phosphatase activity of calcineurin, thereby blocking the transcription of interleukin genes such as IL-2 and reducing cytokine production[3][4]. Daclizumab is a humanized monoclonal antibody targeting CD25, the alpha subunit of the high-affinity interleukin-2 receptor (IL-2R) on activated T cells. By blocking this receptor, daclizumab inhibits IL-2-mediated activation and proliferation of lymphocytes[1][2][6]. The combination was shown to reduce rates of acute transplant rejection when used with corticosteroids. Daclizumab has also been studied for relapsing multiple sclerosis but was withdrawn from clinical use due to safety concerns including severe immune reactions[1][6].
