Overview

Cyclosporine + glucosamine is an investigational combination therapy studied primarily for the treatment of moderate to severe atopic dermatitis. Cyclosporine is a calcineurin inhibitor and immunosuppressant that inhibits T-cell activation by blocking the transcription of interleukin-2 and other cytokines, while glucosamine is an amino sugar with reported immunomodulatory effects, including inhibition of key transcription factors involved in cytokine gene expression and T-cell function. Clinical studies suggest that combining low-dose cyclosporine with glucosamine may enhance therapeutic efficacy compared to cyclosporine alone, leading to greater reductions in disease severity scores (SCORAD index) without increasing adverse events or serum cyclosporine levels. The combination appears to exert additive anti-inflammatory effects, possibly through modulation of chemokines such as CCL17 and reduction in Th2 cytokines like IL-4 and IL-5[1][2][6][8].
