Overview

Cyclosporine + lansoprazole is a combination of two pharmaceutical agents used together in clinical practice. Cyclosporine is an immunosuppressant that acts as a calcineurin inhibitor to suppress T cell activation by inhibiting the phosphatase activity of calcineurin after binding to cyclophilin-1. This prevents the dephosphorylation and activation of nuclear factor of activated T cells (NF-AT), reducing cytokines such as IL-2 and thereby suppressing immune responses[4]. It is primarily indicated for preventing organ transplant rejection and treating certain autoimmune diseases. Lansoprazole is a proton pump inhibitor (PPI) that reduces gastric acid secretion by irreversibly inhibiting the H+,K+-ATPase enzyme in gastric parietal cells[2][5][6]. It is used to treat conditions like peptic ulcers, gastroesophageal reflux disease (GERD), erosive esophagitis, Zollinger-Ellison syndrome, and for Helicobacter pylori eradication when combined with antibiotics[2][5]. When used together or concurrently prescribed in patients with transplants or autoimmune disorders requiring immunosuppression plus gastrointestinal protection or ulcer treatment/prevention, there are clinically significant pharmacokinetic interactions. Lansoprazole can increase blood concentrations of cyclosporine due to shared metabolism via CYP3A4 enzymes; this may raise the risk of adverse effects from elevated cyclosporine levels[3]. Chronic use may also increase risk for hypomagnesemia when both drugs are combined[1].
