Overview

Cyclosporine + semifluorinated alkane (SFA) is a combination drug formulation where the immunosuppressant cyclosporine is dissolved in a semifluorinated alkane vehicle, such as perfluorobutylpentane (F4H5). This novel formulation is designed for topical ophthalmic use, particularly for the treatment of dry eye disease. The SFA serves as a preservative-free, non-stinging carrier that enhances ocular tolerability and may provide faster therapeutic response compared to traditional cyclosporine emulsions. Cyclosporine acts as a calcineurin inhibitor that suppresses T cell activation by inhibiting the phosphatase activity of calcineurin after binding to cyclophilin-1. This prevents dephosphorylation and nuclear translocation of NF-AT transcription factors, reducing cytokines like IL-2 and thereby exerting immunosuppressive effects[1][3][8].
