CYT997 + carboplatin is a combination therapy used in investigational settings for the treatment of recurrent glioblastoma multiforme (GBM). CYT997 is a vascular-disrupting agent (VDA) that targets tumor blood vessels, leading to rapid shutdown of blood flow and subsequent tumor cell death. Carboplatin is a platinum-based chemotherapeutic agent that induces DNA crosslinking, causing DNA damage and apoptosis in rapidly dividing cells. The rationale for combining these drugs is to exploit complementary cytotoxic mechanisms: CYT997 disrupts the tumor vasculature, thereby potentiating the effect of carboplatin-induced DNA damage in tumor cells. This combination has been evaluated in Phase Ib/II clinical trials, demonstrating feasibility and initial signs of activity in relapsed GBM, with ongoing studies to further assess efficacy and safety[1][2][6].
