Drug intelligence / Profile preview
CPX-351 (Vyxeos) is a liposomal formulation of the antineoplastic agents cytarabine and daunorubicin, encapsulated at a fixed 5:1 molar ratio. This specific ratio was identified in vitro to maximize synergistic antileukemic activity. The liposomal delivery system (approximately 100 nm in diameter) protects the drugs from rapid metabolism and excretion, providing a prolonged plasma half-life and preferential accumulation within the bone marrow, where the liposomes are internalized by leukemia cells. CPX-351 is FDA-approved for the treatment of adults and pediatric patients (1 year and older) with newly-diagnosed therapy-related acute myeloid leukemia (t-AML) or AML with myelodysplasia-related changes (AML-MRC). It is also being investigated for efficacy in patients with myelodysplastic syndromes (MDS) and myelodysplastic/myeloproliferative neoplasms (MDS/MPN) who have failed hypomethylating agent therapy.
