Overview

ADxE is a multi-drug chemotherapy regimen used as an induction therapy in the treatment of pediatric acute myeloid leukemia (AML). It was developed as part of the NOPHO-DBH AML 2012 protocol by the Nordic Society of Paediatric Haematology and Oncology (NOPHO) and the Dutch-Belgian-Hong Kong (DBH) consortium. The regimen consists of cytarabine (Ara-C), liposomal daunorubicin (DaunoXome), and etoposide. Cytarabine is a nucleoside analog that interferes with DNA synthesis; liposomal daunorubicin is an anthracycline that inhibits topoisomerase II and intercalates DNA; and etoposide is a podophyllotoxin derivative that also inhibits topoisomerase II. In clinical trials, ADxE is evaluated as a second induction course, often compared against intensified regimens like FLADx to optimize efficacy and minimize toxicity in pediatric patients.
