L-MAG is an induction chemotherapy regimen comprising cytarabine, a liposomal formulation of mitoxantrone, and granulocyte colony-stimulating factor (G-CSF), specifically investigated for the treatment of relapsed or refractory acute myeloid leukemia (AML). The regimen utilizes the improved safety and efficacy profile of liposomal mitoxantrone (PLM60) compared to its conventional form, aiming to reduce cardiotoxicity while maintaining potent antineoplastic activity. G-CSF is included to prime the leukemia cells, potentially increasing their susceptibility to the cytotoxic effects of cytarabine and mitoxantrone by inducing them into the S-phase of the cell cycle, while also supporting hematopoietic recovery. Clinical studies led by The First Affiliated Hospital, Zhejiang University, have evaluated this combination's ability to achieve complete remission in difficult-to-treat AML populations.
