Overview

Cytarabine + thiotepa is a chemotherapy combination used primarily for primary central nervous system lymphoma (PCNSL) and as part of conditioning regimens for stem cell transplantation. Cytarabine is a pyrimidine nucleoside analog that inhibits DNA synthesis, while thiotepa is an alkylating agent that cross-links DNA strands. Both drugs can penetrate the blood-brain barrier, making them effective for CNS malignancies. This combination is often used as part of the MATRix regimen (methotrexate, cytarabine, thiotepa, and rituximab) for PCNSL treatment.
