Overview

A myeloablative conditioning regimen used before allogeneic stem cell transplantation, particularly for patients with acute myeloid leukemia (AML). This combination has demonstrated promising outcomes in cord blood and haploidentical stem cell transplantation. The regimen provides significant anti-leukemic effects with thiotepa and busulfan having good penetration into the central nervous system. Studies have shown that this regimen provides lower relapse rates compared to other conditioning regimens like busulfan plus cyclophosphamide (BuCy), while maintaining similar survival outcomes.
