Cytarabine + venetoclax is a combination regimen used primarily for the treatment of acute myeloid leukemia (AML), especially in adults who are newly diagnosed and ineligible for intensive chemotherapy. Cytarabine is an antimetabolite chemotherapeutic agent that inhibits DNA synthesis by acting as a cytosine analog, leading to cell death in rapidly dividing cells. Venetoclax is a selective small molecule inhibitor of B-cell lymphoma 2 (BCL-2), an anti-apoptotic protein; by inhibiting BCL-2, it promotes apoptosis in malignant hematopoietic cells. The combination has demonstrated improved response rates and overall survival compared to low-dose cytarabine alone in clinical trials such as VIALE-C[1][2][5]. This regimen is approved for use in AML patients who cannot tolerate intensive induction therapy.
