Cytokine-induced killer (CIK) cells are an autologous, ex vivo-expanded population of immune effector cells, primarily characterized by a CD3+CD56+ phenotype. They are generated from a patient's peripheral blood mononuclear cells (PBMCs) through stimulation with interferon-gamma (IFN-γ), interleukin-2 (IL-2), and anti-CD3 antibodies. This cell therapy combines the potent anti-tumor activity of T cells with the non-MHC-restricted cytotoxicity of natural killer (NK) cells. In the context of colon cancer, specifically stage II/III, CIK cells are investigated as an adjuvant immunotherapy following surgical resection and chemotherapy to eliminate residual micrometastases and reduce recurrence rates. The therapy was notably studied in a Phase 2 clinical trial (NCT01800058) led by Yanjuan Zhu, which evaluated the efficacy of CIK cell transfusion in prolonging disease-free survival and overall survival.
