Cytokine-induced memory-like (CIML) natural killer (NK) cells combined with interleukin-2 (IL-2) is an investigational cellular immunotherapy regimen being evaluated by the Dana-Farber Cancer Institute for the treatment of recurrent, high-grade ovarian cancer. The therapy involves the administration of autologous NK cells that have been ex vivo pre-activated with a cytokine cocktail (typically IL-12, IL-15, and IL-18) to induce a memory-like state characterized by enhanced proliferation, longevity, and potent anti-tumor cytotoxicity. These CIML NK cells are delivered via intraperitoneal infusion to directly target the abdominal environment common in ovarian cancer progression. Following cell transfer, patients receive low-dose subcutaneous IL-2 to promote the expansion and sustained activity of the infused NK cells. This approach aims to overcome the immunosuppressive tumor microenvironment and provide a durable immune response against platinum-resistant ovarian malignancies, including high-grade serous, endometrioid, and clear cell carcinomas.
