Overview
d-cycloserine + acamprosate is a small molecule combination therapy being developed by Confluence Pharmaceuticals for the treatment of Fragile X Syndrome and alcohol use disorders. The combination consists of d-cycloserine (DCS), a partial agonist at the glycine site of the N-methyl-D-aspartate (NMDA) receptor, and acamprosate (ACP), which modulates glutamatergic and GABAergic neurotransmission. By targeting glutamatergic dysfunction—a key feature of both Fragile X Syndrome and alcohol dependence—the therapy aims to improve neurological and cognitive outcomes. Confluence Pharmaceuticals is advancing this combination alongside standalone formulations of acamprosate, leveraging the complementary mechanisms of both drugs to address neurological and cognitive conditions.
