D3S-001 + cisplatin is a combination therapy being developed by D3 Bio (Wuxi) Co., Ltd for the treatment of advanced solid tumors harboring the KRAS G12C mutation. D3S-001 is a potent, selective, and covalent small molecule inhibitor of the KRAS G12C mutant protein, designed to lock the protein in its inactive GDP-bound state and disrupt downstream oncogenic signaling. Cisplatin is a platinum-based chemotherapy agent that acts by forming covalent cross-links with DNA, primarily at the N7 position of purine bases, which interferes with DNA replication and transcription, ultimately leading to cell death. This combination is being evaluated in Phase 1/2 clinical trials (e.g., NCT05162430) to assess its safety and efficacy in patients with KRAS G12C-mutated cancers, including non-small cell lung cancer (NSCLC) and colorectal cancer (CRC). D3S-001 has received Breakthrough Therapy Designation from the FDA for the treatment of KRAS G12C-mutated NSCLC.
