Overview

This is an **experimental combination therapy** consisting of three components: dabrafenib (a selective BRAF inhibitor), LTT462 (a selective ERK1/2 inhibitor), and spartalizumab (a monoclonal antibody targeting the programmed cell death protein 1, PD-1). This triplet targets the MAPK pathway at two levels (BRAF and ERK) and adds immunotherapy via PD-1 blockade. The rationale is to overcome resistance to single-targeted therapies and synergistically enhance anti-tumor immune responses. Early-phase clinical trials are investigating this combination in adult patients with advanced solid tumors including BRAF V600-mutant cancers[5][6][8].
