Overview

Dabrafenib and trametinib are small molecule kinase inhibitors used in combination as a targeted therapy for cancers with BRAF V600 mutations. Dabrafenib is a selective inhibitor of the BRAF protein, which is part of the MAPK/ERK signaling pathway that promotes cell proliferation and survival. Trametinib inhibits MEK1 and MEK2, downstream kinases in the same pathway. By blocking both BRAF and MEK, this combination disrupts cancer cell growth more effectively than either agent alone. The combination is primarily indicated for melanoma (including adjuvant treatment after surgery), advanced non-small cell lung cancer (NSCLC), certain types of thyroid cancer, solid tumors with BRAF V600E mutation in adults and children 6 years or older, and glioma in children 1 year or older.
