Overview

The combination of **dabrafenib**, **trametinib**, and **panitumumab** is an investigational regimen targeting advanced or metastatic BRAF V600E-mutant colorectal cancer. Dabrafenib is a small-molecule inhibitor of mutant BRAF kinase, trametinib is a small-molecule inhibitor of MEK1/2 (part of the MAPK pathway), and panitumumab is a monoclonal antibody targeting the epidermal growth factor receptor (EGFR). The rationale for this triplet therapy is to block compensatory feedback reactivation of the MAPK pathway in BRAF-mutant colorectal tumors that are resistant to BRAF inhibition alone. This combination has been studied in phase I/II trials, demonstrating antitumor activity and a safety profile characterized chiefly by dermatologic, gastrointestinal, and constitutional side effects[1][3][8].
