Overview

This combination chemotherapy regimen consists of three cytotoxic agents—dacarbazine, doxorubicin, and ifosfamide—used primarily for the treatment of advanced or metastatic soft tissue sarcomas. The regimen is notable for its high response rates compared to single-agent or two-drug combinations in this setting. Doxorubicin intercalates into DNA and inhibits topoisomerase II; ifosfamide is an alkylating agent that cross-links DNA strands; dacarbazine also acts as an alkylating agent and antimetabolite.
