Overview
dacarbazine + Endostar is a combination therapy consisting of the recombinant human endostatin (Endostar) and the alkylating chemotherapy agent dacarbazine. Endostar, developed by Simcere Pharmaceutical, is a potent endogenous angiogenesis inhibitor that targets tumor vascularization by inhibiting endothelial cell proliferation, migration, and tube formation. Dacarbazine is a small molecule alkylating agent that has been a standard treatment for metastatic melanoma, working by causing DNA damage to inhibit tumor cell division. The combination was evaluated in a Phase II trial for untreated metastatic melanoma without c-kit or BRAF mutations, showing significant improvements in progression-free survival and overall survival compared to dacarbazine alone. While Endostar is approved in China for non-small cell lung cancer, its use in combination with dacarbazine for melanoma remains in clinical development.
