Overview

Dacarbazine + temozolomide refers to a chemotherapy regimen utilized in clinical research, specifically within a Phase II trial (NCT05162833) sponsored by The First Hospital of Jilin University for the adjuvant treatment of PD-L1-positive, resectable mucosal melanoma. In this clinical context, the regimen serves as a chemotherapy backbone where patients receive either dacarbazine or temozolomide, often in combination with cisplatin. Both dacarbazine and temozolomide are small molecule alkylating agents that act as prodrugs, undergoing conversion to the active metabolite 5-(3-methyltriazen-1-yl)-imidazole-4-carboxamide (MTIC). MTIC exerts its cytotoxic effect by methylating DNA at the O6 and N7 positions of guanine, which results in DNA strand breaks, inhibition of DNA replication, and the induction of apoptosis. While typically used as alternatives due to their identical active metabolite, they are grouped together in this study's protocol to provide flexible chemotherapy options for mucosal melanoma patients.
