Overview

A combination therapy consisting of **dacetuzumab** (a humanized monoclonal antibody targeting CD40), **lenalidomide** (an immunomodulatory small molecule), and **dexamethasone** (a synthetic glucocorticoid corticosteroid), used or investigated primarily for treating multiple myeloma. Dacetuzumab exerts antitumor effects via direct apoptotic signaling through CD40 as well as Fc-mediated effector cell functions (ADCC/ADCP). Lenalidomide modulates the immune system and directly inhibits myeloma cells by affecting cytokine production and T-cell/NK-cell activation. Dexamethasone, a potent anti-inflammatory and immunosuppressant, is widely used in myeloma regimens to induce apoptosis in lymphoid cells and decrease treatment-related toxicity, such as infusion reactions. The combination has shown preliminary evidence of synergistic anti-myeloma effects in vitro and was evaluated in trials for relapsed/refractory multiple myeloma[3][5].
