Overview

The combination of **daclizumab** and **corticosteroids** is an immunosuppressive regimen primarily studied in the context of organ transplantation. Daclizumab is a humanized monoclonal antibody that targets the alpha subunit (CD25) of the interleukin-2 (IL-2) receptor on activated T cells, competitively inhibiting IL-2-mediated T-cell proliferation. Corticosteroids (such as prednisone or methylprednisolone) provide broad immunosuppression by acting as agonists at the glucocorticoid receptor, thereby modulating gene expression to reduce the production of inflammatory cytokines. This specific combination was evaluated by **Astellas Pharma** in the Phase 2 CHASE trial (NCT00295607) for liver transplant recipients with Hepatitis C (HCV) to assess the impact of steroid-free or steroid-reduced regimens on viral recurrence and graft rejection. Daclizumab (Zenapax) was originally developed by **Roche** but has since been discontinued for transplant indications.
