Dacomitinib + anlotinib is a combination therapy consisting of two tyrosine kinase inhibitors. Dacomitinib is a second-generation irreversible EGFR tyrosine kinase inhibitor that specifically targets EGFR mutations, particularly exon 19 deletion or exon 21 L858R substitution mutations. It works by irreversibly binding to the ATP domain of the EGFR family kinase domains. Anlotinib is a novel multi-targeting tyrosine kinase inhibitor that targets several receptor tyrosine kinases (RTKs). It's currently authorized as a third-line treatment for patients with advanced NSCLC. This combination is being investigated for the treatment of non-small cell lung cancer (NSCLC), particularly as a first-line treatment option. When combined, these drugs may provide enhanced efficacy by simultaneously inhibiting multiple cancer growth pathways.
