Overview

A combination therapy consisting of dactolisib (BEZ235), an oral dual inhibitor of class I PI3K and mTOR complexes 1 and 2, and trastuzumab, a monoclonal antibody targeting HER2. This combination is primarily studied for HER2-positive advanced breast cancer, particularly in patients who have developed resistance to trastuzumab alone. The combination targets both the PI3K/mTOR pathway and the HER2 receptor to overcome resistance mechanisms.
