Overview

JTT-705 + pravastatin is an investigational combination regimen consisting of the cholesteryl ester transfer protein (CETP) inhibitor dalcetrapib (JTT-705) and the HMG-CoA reductase inhibitor pravastatin. Developed by Akros Pharma (a subsidiary of Japan Tobacco), the regimen was designed to treat Type II hyperlipidemia by simultaneously raising high-density lipoprotein cholesterol (HDL-C) and lowering low-density lipoprotein cholesterol (LDL-C). Dalcetrapib works by inhibiting CETP, which normally facilitates the transfer of cholesteryl esters from HDL to LDL and VLDL; its inhibition leads to increased HDL levels. Pravastatin, a statin, reduces cholesterol synthesis in the liver. While Phase II trials demonstrated significant increases in HDL-C and reductions in CETP activity, the development of dalcetrapib was eventually halted by Roche after Phase III trials (dal-OUTCOMES) failed to show a reduction in cardiovascular events.
