Dalpiciclib + abiraterone + prednisone is a combination therapy under investigation for the treatment of prostate cancer, particularly metastatic castration-resistant prostate cancer and high-risk non-metastatic prostate cancer. Dalpiciclib is a selective cyclin-dependent kinase 4/6 inhibitor that blocks cell cycle progression, thereby inhibiting tumor cell proliferation. Abiraterone acetate is an androgen biosynthesis inhibitor that works by irreversibly inhibiting Cytochrome P450 17A1 (CYP17A1), an enzyme critical to androgen production; it reduces androgen levels and thus limits the growth stimulus for prostate tumors. Prednisone is a synthetic glucocorticoid used to counteract mineralocorticoid excess caused by CYP17A1 inhibition with abiraterone and provides glucocorticoid replacement therapy. This combination aims to intensify treatment efficacy in advanced or high-risk forms of prostate cancer[1][2][3][8].
