Overview

This combination therapy consists of dalpiciclib (SHR6390) and SHR7390, two small molecule inhibitors developed by Jiangsu Hengrui Pharmaceuticals. Dalpiciclib is a selective cyclin-dependent kinase 4/6 (CDK4/6) inhibitor that was approved in China in 2021 for the treatment of breast cancer; it works by blocking the transition of cells from the G1 to the S phase of the cell cycle. SHR7390 is an investigational inhibitor of mitogen-activated protein kinase kinase (MEK), a key component of the RAS/RAF/MEK/ERK signaling pathway often dysregulated in various cancers. The combination is currently being evaluated in a Phase I clinical trial sponsored by Henan Cancer Hospital to assess its safety and tolerability in patients with metastatic digestive system tumors, a population where dual inhibition of the cell cycle and MAPK signaling may overcome resistance to monotherapy.
