Dalutrafusp alfa + mFOLFOX6 is a combination regimen consisting of dalutrafusp alfa, an investigational bifunctional fusion protein targeting CD73 and TGF-beta, and the mFOLFOX6 chemotherapy regimen. mFOLFOX6 includes oxaliplatin, leucovorin (folinic acid), and fluorouracil (5-FU), and is used as a standard chemotherapy backbone for gastrointestinal malignancies such as colorectal cancer. Dalutrafusp alfa is developed to enhance antitumor immune response by blocking the immunosuppressive effects of TGF-beta and adenosine signaling (via CD73), while mFOLFOX6 acts by inhibiting DNA synthesis and repair and interfering with folate metabolism to induce cytotoxicity in rapidly dividing cancer cells.
