danazol + rhTPO

Danazol + rhTPO is a combination therapy consisting of danazol, a synthetic steroid with androgenic and anti-gonadotropic properties, and recombinant human thrombopoietin (rhTPO), a biologic that stimulates platelet production. This combination was studied in a multicenter randomized controlled trial for the treatment of persistent primary immune thrombocytopenia (ITP) in patients who had failed glucocorticosteroid treatment. In the trial, the rhTPO + danazol combination demonstrated significantly higher response rates compared to danazol alone, with major response rates of 38.4% versus 7.9% and total response rates of 60.3% versus 36.5%. The combination therapy markedly increased platelet counts in chronic ITP patients and was generally well tolerated with mild adverse events. Danazol is a small molecule drug, while rhTPO is a biologic (recombinant protein). The biological target of rhTPO is the thrombopoietin receptor, which stimulates megakaryocyte production and platelet formation.
