Dapagliflozin + gliclazide is a fixed-dose combination (FDC) therapy developed by Servier for the treatment of type 2 diabetes mellitus (T2DM). It combines two small-molecule antidiabetic agents with complementary mechanisms of action: dapagliflozin, a selective sodium-glucose cotransporter-2 (SGLT2) inhibitor, and gliclazide, a second-generation sulfonylurea. Dapagliflozin works by blocking the reabsorption of glucose in the proximal tubule of the kidney, thereby increasing urinary glucose excretion and lowering blood sugar levels independently of insulin. Gliclazide stimulates insulin secretion from the pancreatic beta cells by binding to the sulfonylurea receptor 1 (SUR1) and closing ATP-sensitive potassium channels, which leads to cell depolarization and insulin release. The combination is typically formulated as a sustained-release (SR) tablet (e.g., 10 mg dapagliflozin and 60 mg gliclazide SR) and has been evaluated in Phase III clinical trials for patients whose glucose levels are inadequately controlled by metformin monotherapy.
