Overview

Dapagliflozin + glimepiride is a fixed-dose combination of two oral antidiabetic agents used in the management of type 2 diabetes mellitus. Dapagliflozin is a sodium-glucose co-transporter 2 (SGLT2) inhibitor that lowers blood glucose by inhibiting SGLT2 in the proximal renal tubules, reducing glucose reabsorption and increasing urinary glucose excretion. Glimepiride is a second-generation sulfonylurea that stimulates insulin secretion from pancreatic beta cells by blocking ATP-sensitive potassium channels, leading to depolarization and subsequent insulin release. The combination improves glycemic control through complementary mechanisms—dapagliflozin acts independently of insulin, while glimepiride enhances endogenous insulin secretion[3][4][6]. This therapy is primarily indicated for adults with type 2 diabetes inadequately controlled on monotherapy or as part of combination regimens.
