Dapsone + adapalene is a topical combination therapy used primarily for the treatment of acne. Dapsone is a sulfone antibiotic that works by inhibiting bacterial synthesis of dihydrofolic acid through competitive inhibition of dihydropteroate synthase, thereby reducing bacterial growth and inflammation. It also exhibits anti-inflammatory effects by inhibiting myeloperoxidase activity in neutrophils, which reduces tissue damage during inflammation[1][4][6][8]. Adapalene is a synthetic retinoid (a derivative of vitamin A) that acts as a modulator of cellular differentiation, keratinization, and inflammatory processes. It works by loosening cells on the skin’s surface and unblocking pores through reduced oil production, thus decreasing pimples, whiteheads, and blackheads[5]. The combination leverages both antibacterial/anti-inflammatory actions (dapsone) and comedolytic/keratolytic effects (adapalene), making it effective for mild to moderate acne vulgaris[5].
